چکیده
Cow milk is an important source of food protein for children, however, it could lead to allergy, especially infant. α-lactalbumin (α-LA) and β-lactoglobulin (β-LG) from whey make up a relatively high proportion proteins have received widespread attention as major allergens in milk. However, few studies identified the epitopes both simultaneously. In this study, ImmunoCAP indirect ELISA were first used detection sIgE screen sera allergic patients with binding capacity α-LA β-LG. Subsequently, mimotopes was biopanned by phage display technology bioinformatics 17 mimic peptide sequences obtained. Aligned or β-LG, we one linear epitope on at AA11–26 5 β-LG AA 9–29, 45–57, 77–80, 98–101, 121–135, respectively. Meanwhile, 8 conformational their distributions located using Pepitope Server espectively. Finally, Glutamine lysine determined common residues Moreover, found addition mouse antihuman IgE during biopanning process did not significantly affect identification epitopes.

Isolation and purification of beta-lactoglobulin from cow milk
AIM The present study was undertaken to standardize a convenient method for isolation and purification of β-lactoglobulin (β-lg) from cow milk keeping its antigenicity intact, so that the purified β-lg can be used for detection of cow milk protein intolerance (CMPI). MATERIALS AND METHODS Raw milk was collected from Gir breed of cattle reared in Haringhata Farm, West Bengal. Milk was then con...
متن کاملPuzzling Peptides from a Phage Display Library
The commercial availability of random peptide libraries displayed on the M13 phage is increasing their use forstudies on epitope identification, enzyme inhibitors, receptor ligands, etc. In this study two experimentswhere planned for selection of peptides. First with sheep antibodies, the positive selector was IgG, preparedon Protein G column from a pool of 11 sheeps immunized...
متن کاملβ-lactoglobulin and α-lactalbumin hydrolysates as sources of antibacterial peptides
the presence of antibacterial activity in bovine β-lactoglobulin and in α-lactalbumin hydrolysates was investigated. the plasmin-digest of β-lactoglobulin (pdβ) and of α-lactalbumin (pdα) were fractionated, using reversed phase high performance liquid chromatography. the antibacterial activity of β-lactoglobulin, α-lactalbumin, nisin, plasmin, pdβ and pdα were in vitro tested against pathogenic...
متن کاملConformational epitopes on the diabetes autoantigen GAD65 identified by peptide phage display and molecular modeling.
The major diabetes autoantigen, glutamic acid decarboxylase (GAD65), contains a region of sequence similarity, including six identical residues PEVKEK, to the P2C protein of coxsackie B virus, suggesting that cross-reactivity between coxsackie B virus and GAD65 can initiate autoimmune diabetes. We used the human islet cell mAbs MICA3 and MICA4 to identify the Ab epitopes of GAD65 by screening p...
متن کاملDetection and Quantifi cation of αS1-, αS2-, β-, κ-casein, α-lactalbumin, β-lactoglobulin and Lactoferrin in Bovine Milk by Reverse-Phase High- Performance Liquid Chromatography
Bovine milk proteins has been widely studied because of the strong association and relationship with composition and technological properties of milk. Cow’s milk quality is very important, above all in such countries like Italy, where about 70% of whole milk production is used in cheese-making industry. A reversed-phase highperformance liquid chromatography (RP-HPLC) method was developed to ide...
