Glutamine and lysine as common residues from epitopes on α-lactalbumin and β-lactoglobulin from cow milk identified by phage display technology

Cow milk is an important source of food protein for children, however, it could lead to allergy, especially infant. α-lactalbumin (α-LA) and β-lactoglobulin (β-LG) from whey make up a relatively high proportion proteins have received widespread attention as major allergens in milk. However, few studies identified the epitopes both simultaneously. In this study, ImmunoCAP indirect ELISA were first used detection sIgE screen sera allergic patients with binding capacity α-LA β-LG. Subsequently, mimotopes was biopanned by phage display technology bioinformatics 17 mimic peptide sequences obtained. Aligned or β-LG, we one linear epitope on at AA11–26 5 β-LG AA 9–29, 45–57, 77–80, 98–101, 121–135, respectively. Meanwhile, 8 conformational their distributions located using Pepitope Server espectively. Finally, Glutamine lysine determined common residues Moreover, found addition mouse antihuman IgE during biopanning process did not significantly affect identification epitopes.
